Enveda reports positive early safety data for ENV-308, an experimental oral drug inspired by exercise-induced molecule Lac-Phe, as it advances toward larger studies to assess its potential in weight regain prevention after GLP-1 therapy.
Enveda has reported encouraging first human data for ENV-308, an experimental oral treatment designed to mimic Lac-Phe, a molecule the body produces during intense exercise and that has drawn attention in obesity research. According to the company, the Phase I study involved 88 healthy adults and was intended to assess safety, tolerability and how the drug behaves in the body. Enveda said the compound was well tolerated across the dose range tested and produced an unusually strong gastrointestinal safety profile. That matters because nausea, vomiting and related side effects are among the main reasons people stop taking GLP-1 medicines.
The company said there were no serious adverse events, no treatment discontinuations and no dose interruptions in the trial. Independent summaries of the results described the findings as favourable, while noting that the study was not designed to test whether ENV-308 causes weight loss in patients. Still, the early signal has raised interest because the drug is being positioned as a possible follow-on treatment for people who stop GLP-1 therapy and later regain weight.
Enveda is also pointing to an exploratory drop in leptin, a hormone involved in appetite regulation, as a sign that the medicine is affecting pathways linked to metabolism. The company said that this finding echoes animal studies in which ENV-308 helped preserve lean muscle during weight loss and reduced weight regain after treatment ended. Those preclinical results will not predict clinical benefit on their own, but they help explain why the programme has attracted attention.
The next step is likely to be larger studies, including a planned Phase II trial, which Enveda says will look at whether ENV-308 can help maintain weight loss and metabolic gains after GLP-1 drugs are stopped. The company, based in Boulder, Colorado, says the drug was discovered using its artificial intelligence platform PRISM, which it uses to search for biologically active molecules found in nature. For now, the latest data suggest the candidate has cleared an important early safety hurdle, but its effectiveness in people with obesity remains unproven.
